1.Intended Use
Phoenix Virus NAT® is a fast, simple and inexpensive method for preparing one to hundreds of samples for RT-PCR amplification. It allows viral nucleic acid preparation from nasopharyngeal aspirate and lavage, bronchoalveolar lavage, saliva, nasopharyngeal swab, oropharyngeal swab, and sputum samples in VTM or UTM. Highly denaturating transport media containing GTC (e.g., Phoenix Protect, Zymo DNA/RNA Shield etc) are not compatible for the fast extraction. After incubation of 5 minutes at room temperature the sample can be added directly to the PCR mix.
2.Storage of clinical specimens
Clinical specimens diluted in Phoenix Virus NAT® can be stored at 2-8°C for up to 5 days.
3.Materials Required
- 10-100 µl micropipette and compatible tips (DNase and RNase free)
- 5- or 2-ml microcentrifuge tubes
- Powder-free nitrile disposable gloves
4.Application Protocol
- Take 75 µl of Phoenix Virus NAT® to a clean microcentrifuge tube.
- Add 25 µl of VTM / UTM sample and mix by pipetting 4-5 times.
- Incubate at room temperature for 5 minutes.
- Sample can be used directly for PCR reaction or stored at 2-8°C.

The use of PhoenixDx® SARS-CoV-2 detection kits and PhoenixDx® SARS-CoV-2 Mutant Screen kits in combination with Phoenix Virus NAT® is recommended.
